Dark current — the thermally generated charge that accumulates in each pixel in the absence of illumination — is one of the most important noise sources in scientific imaging. Step 3: Define the spectral range. Silicon sensors cover 200–1100 nm; InGaAs covers 0.9–1.7 µm (or 2.5 µm extended); InSb and MCT cover the mid- and long-wave infrared. Step 4: Define the spatial resolution and field of view, which together with the optics determine the required pixel count and pixel size. Step 5: Define the cooling, interface, triggering, and software requirements based on the experimental setup [1, 3, 5].
10.2Application-Specific Recommendations
For widefield fluorescence microscopy, the default recommendation is a back-illuminated sCMOS camera with 1–2 e⁻ read noise, 80–95% peak QE, and 30–100 fps frame rate. The large field of view (typically 13 × 13 mm sensor, corresponding to 220 × 220 µm at 60× magnification) and high frame rate enable calcium imaging, live-cell time-lapse, and high-content screening with excellent sensitivity. For single-molecule and super-resolution microscopy requiring the absolute lowest noise, an EMCCD with deep cooling remains the established choice, although qCMOS cameras are rapidly closing the gap [1, 9].
For spectroscopy (Raman, fluorescence, absorption), a deep-cooled back-illuminated CCD with large pixels (13–26 µm), high QE (> 90% at peak), and ultra-low dark current (< 0.001 e⁻/pixel/s at −80 °C) provides the best combination of sensitivity and dynamic range. FVB and on-chip binning should be available for maximum SNR with faint spectra. For high-speed spectral acquisition, a back-illuminated sCMOS with fast readout and low read noise is preferred. For astronomy, large-format back-illuminated CCDs or CMOS sensors with deep cooling and low read noise are the standard; the choice between CCD and CMOS depends on the telescope's readout speed requirements and the availability of mosaic focal-plane assemblies. For high-speed imaging (> 1000 fps), global-shutter CMOS cameras with on-board memory provide the combination of speed and image quality required for transient event capture.
